AB0170 ACTIVATED SENESCENCE PATHWAYS IN THE TEMPORAL ARTERIES OF PATIENTS WITH GIANT CELL ARTERITIS

چکیده
Background Giant cell arteritis (GCA) is an inflammatory disease marked by granulomatous inflammation of the large- and medium-vessels. GCA exclusively affects older patients above 50 years old, suggesting a role senescence in pathobiology GCA. IL-6 receptor blocking therapy has been shown to be effective treatment for GCA.(1) More recently, phase two randomized control trial efficacy GM-CSF GCA.(2) Both are senescence-associated secretory phenotype (SASP) cytokines,(3) implicating roles cellular SASP immunopathology Objectives We seek find clues senescent inflamed arteries patients. Methods Immunohistochemical stainings markers p21 WAF1/CIP1 p16/INK4A as well SAPS cytokines were performed on temporal artery biopsies (TABs; n=8) from with giant arteritis. Non-inflamed age-matched controls included (n=14). Stainings scored quantitatively using QuPath software. Opal based multiplex fluorescent confirm colocalization GM-CSF. Results All four detected arteries, most notably cells surrounding intimal lamina elastica although expression other was also throughout vessels. The all significantly lower arteries. Cells positive that co-express Conclusion Our data suggest presence activated pathways at site vascular support further research into References [1]Maz M, Chung SA, Abril A, Langford CA, Gorelik Guyatt G, Archer AM, Conn DL, Full KA, Grayson PC, et al. 2021 American College Rheumatology/Vasculitis Foundation Guideline Management Cell Arteritis Takayasu Arteritis. Increased angiotensin II type 1 receptor expression in temporal arteries from patients with giant cell arteritis.
PURPOSE Currently, giant cell arteritis (GCA) is primarily treated with corticosteroids or immunomodulating agents, but there is interest in identifying other noncorticosteroid alternatives. Similarities exist in the injury pathways between GCA and atherosclerosis. Angiotensin II is a vasoactive peptide involved in vessel inflammation during atherosclerosis, and angiotensin II receptor inhibito...
متن کاملRelapses in Patients With Giant Cell Arteritis
Giant cell arteritis (GCA) is a relapsing disease. However, the nature, chronology, therapeutic impact, and clinical consequences of relapses have been scarcely addressed. We conducted the present study to investigate the prevalence, timing, and characteristics of relapses in patients with GCA and to analyze whether a relapsing course is associated with disease-related complications, increased ...
متن کاملElectron microscopy of giant-cell (temporal) arteritis.
Definitive histological studies of giant-cell (temporal) arteritis have been published by several workers (Harrison, 1948; Heptinstall, Porter, and Barkley, 1954; McCormick and Neuberger, 1958). The lesions are fairly uniform and have involved all layers of many different arteries and veins-large and small, intracranial as well as extracranial. The distinctive histological finding in this disea...
متن کاملGiant cell arteritis presenting with bilateral orbital inflammatory disease and enhancing superficial temporal arteries
To cite: Mitchell JR, KrashinBichler I, Rosenblum M, et al. Pract Neurol 2014;14: 446–447. CASE REPORT A 68-year-old man with metastatic prostate cancer was admitted with a 1-week history of bilateral ‘conjunctivitis’, headaches and horizontal diplopia. On examination, the superficial temporal arteries were bilaterally prominent and indurated, with conjunctival injection and chemosis. There wer...
متن کاملTakayasu's arteritis and giant cell (temporal or cranial) arteritis.
Takayasu's arteritis (TA), of which the first case was described by Takayasu in 1905 (1), is a chronic inflammatory disease of unknown etiology that primarily involves the aorta, its major branches, and the pulmonary and coronary arteries. Synonymssuch as aortic arch syndrome, pulseless disease and aortitis syndrome reflect this anatomic localization. TAappears to be most commonin East Asia inc...
